GENETIC TRANSFORMATION OF Brassica napus L. HANNA VARIETY WITH THE HELP OF Agrobacterium tumefaciens AGLO

The authors investigated the effect of hormonal composition of media and concentration of hygromicin antibiotic on regeneration of sprouts from cotyledonous explants of Brassica napus L. Hanna variety. For isolation of transgenic plants the authors use the strain of Agrobacterium tumefaciens AGL0 with plasmid pCAMBIA 1305.1, containing the lectin genes of pea (psl), β-glucuro-nidase (gus) and hygromicin phosphotransferase (hptII) in the T-DNA. During using of hygromicin as selective marker, the efficiency of transformation reaches to 1,8 %. PCR-analysis of DNA from transgenic plants confirms the presence and the expression of translocated gene of psl on mRNA level.
